چکیده

Hydroxyurea is thought to inhibit DNA synthesis but the site of inhibition has not been established. The urinary excretion of orotic acid was measured in 6 patients, 5 with acute leukemia and 1 with metastatic melanoma, receiving 6-azauridine which is known to inhibit orotidylic decarboxylase. The orotic aciduria observed in these patients was reversed by the addition of hydroxyurea, suggesting inhibition of de novo pyrimidine biosyn thesis at a step prior to the formation of orotic acid. No clear-cut potentiation of a chemotherapeutic effect of this combination of drugs was observed.

Orotic Acid, Nucleotide-Pool Imbalance, and Liver-Tumor Promotion: A Possible Mechanism for the Mitoinhibitory Effects of Orotic Acid in Isolated Rat Hepatocytes1

This study was designed to determine the possible mechanism by which orotic acid exerts its mitoinhibitory effect on rat hepatocytes in primary culture. Orotic acid inhibited, dose-dependently DNA synthesis in hepatocytes induced by epidermal growth factor, transforming growth factor a, hepatocyte growth factor, acidic fibroblast growth factor, or plasma from rats exposed to various liver cell-...

Side Effects of Hydroxyurea in Patients with Sickle Cell Anemia

Background: Hemoglobin S arises is the result of a point mutation (A-T) in the sixth codon on the -globin gene on chromosome 11 causing sickle cell anemia. The presence of fetal hemoglobin in infancy plays a relatively protective role for vaso-occlusive symptoms that are the major contributor for the morbidity and mortality among patients with sickle cell anemia. hydroxyurea, an s-phase-specif...

The relationship between urea and pyrimidine de novo synthesis in ruminant liver.

The influence of ammonia and inhibition of the urea cycle by norvaline on orotic acid synthesis in five Polish Merino ewes was investigated. The sheep were exposed to 60 min of successive infusions of 0.9% NaCl, NH4Cl (20 mumol kg-1 min-1), and NH4Cl with DL-norvaline (10 mumol kg-1 min-1) solutions administered to the mesenteric vein. The average rate of orotic acid output, urea output and amm...
